Pisonia grandis R. Br. is extensively used as leafy vegetable and in Indian traditional medicine as an anti-diabetic and for other medicinal purposes. The leaf extracts were reported to posjses numerous bioactive compounds includes pinitol, allantoin,octacosanol, p-sitosterol, a- spinasterol, p-sitosterol, p-D-glucopyranoside, dulcitol, flavonoids and quercetin. The cells under culture accumulate large amounts of secondary compounds under specific culture conditions. Therefore, in vitro propagationof medicinal plant can be used as fast and reliable method to synthesis the bioactive compounds. Moreover; P. grandis is reproduced by vegetative means and success rate of production of viable sapling is also very low. Besides; young saplings are affected with leaf rust. Therefore; effective strategies to be developed for the production of viable saplings of this multipurpose plant species by using plant tissue culture technique. The present study describes an effective; reproducible and rapid regeneration protocol for in vitro shoot development of P. grandis achieved through shoot tip and nodal explants on Murashige and Skoog's medium. Maximum shoots (17.5±0.70) per explants were obtained on MS medium supplemented with 1.0 mg/1 BAP along with 0.5 mg/1 NAA. In vitro regenerated shoots rooted best on half-strength MS medium containing 1.0 mg/1 of IBA. Rooted shoots were successfully transferred to field conditions with 75% survival.
