An efficient method for plant regeneration of Asparagus officinalis through somatic embryogenesis was developed. Callus was obtained from seedling segments on Murashige and Skoog (MS) medium supplemented with 1 mg/l naphthalene acetic acid (NAA) + 0.5 mg/l benzyleadenine (BA). The highest frequency of embryogenic callus and then conversion to embryos were observed with a medium containing 1 mg/l 2,4- dichlorophenoxy acetic acid (2,4-D) + 1 mg/l kinetin (kin). The highest percentage of embryo germination was obtained when the medium contained 0.5 mg/l 2,4-D + 0.5 mg/l kin was used. Among three concentrations of both adenine sulfate and casien hydrolysate added sperately to the germination medium; 80 mg/l of adenine sulfate obviously enhanced shootand root length. However, the highest percentage of embryo germination and highest number of roots were recorded when 2000 mg/l casien hydrolysate was added to the germination medium. The highest percentage of survival was obtained when plantlets in sizeof 5-10 cm shoot to 3-5 cm root were transplanted in peat moss and parlite (1:1).
