Glucose metabolism in erythrocytes obtained from buffaloes with phosphorus deficiency haemoglobinuria was studied in vitro. The glucose consumption and lactate production rates in affected erythrocytes were similar to normal erythrocytes. In affected erythrocytes, inorganic phosphorus (Pi), ATP, acid-labile and organic phosphates were significantly depleted. When these erythrocytes were incubated in medium containing Pi, the cellular phosphates were replenished. After administration of sodium acidphosphate i.v. to the affected buffaloes, the erythrocyte ATP levels increased rapidly and in many cases the clinical recovery corroborated with increases in cellular phosphates. It is suggested that with phosphorus deficiency haemoglobinuria the increased haemolysis is due to impaired shape transformation in the ATP-deficient erythrocytes. The possible mechanisms of haemolysis are discussed.
