The superoxide anion (O_~- ) generation in rabbit neutrophils was examined under exposure to diesel exhaust matter (DEM) or bisulfite (HSO_3~-) in vitro. Nertrophils were adjusted to 2 x 10~6 cells/ml for the reaction. The reactions were repeated using neutrophils from 2 rabbits. In the prsence of phorbolmyristate acetate (PMA), DEM (0.011 mg/ml) caused 0.38 nmol/10~6 cells/ min of superoxide anion generation, which was slightly lower than that caused by the exposure of 41 mM bisulfite. Although higher bisulfites (82 mM and 165 mM) caused greater superoxide anion generations (0.62 and 0.64 nmol/10~6 cells/min, respectively), it was presumed that they would have also reduced cytochrome c, which is used for superoxide anion dismutation. In the presence of zymosan, reactions of the anion generation gradually proceeded and continued during the whole observation period. It directly affected neutrophils to produce superoxide anions.
