This study was conducted to determine the organization of nuclear pore complex (NPC) in the nuclei of germinal vesicle (GV) isolated from immature oocytes. Isolated oocytes for medium antral follicles and preovulatory follicles of prepubertal gilts were synchronized and matured in vitro. Results revealed that an average of 66 NPC/1 micro m2 was detected in the GV of isolated antral follicles with an average diameter of 82+or-6.53 nm. Whole oocytes from preovulatory follicles and in vivo matured oocytes showed a stable arrangement of the NPC on the GV with 66 and 65 NPC/1 micro m2, respectively, at the resumption of meiotic maturation. Functional studies are in progress in order to develop an experimental model able to modulate the arrangement of NPC, during the first phase of meiotic resumption, in order to correlate the arrangement of NPCs with the developmental potential of the fully-grown oocytes..
