The study was conducted to determine the effect of different culture conditions on type 5 capsular polysaccharide production of Staphylococcus aureus isolated from dairy cattle. Type 5 capsular polysaccharide was cultured in BHI, solid columbia, and mod 110 culture media. Glucose and lactose were utilized as carbon sources in every culture in solid and liquid states. Results revealed that capsular polysaccharide production decreased in BHI culture media, but increased in mod 110. It was observed that solid culture media had better capsular polysaccharide production, while lactose as carbon source enhanced capsular polysaccharide production.
