Effect of Fraxinus excelsior on dermal collagen during wound healing in adult Oryctolagus caniculus rabbits.

Effect of Fraxinus excelsior alcoholic extract on thickness of dermal papillary collagen during healing in 4 male and 3 female adult Oryctolagus caniculus rabbits is described. Under anesthesia, 4 wounds were created on each rabbit and treated daily with 0.2g eucerin as negative controls, phenytoin 1% positive controls and Fraxinus excelsior 10%, and 20% in eucerin-based creams for 20 days. At euthanasia on day 21, skin sections were taken and stained with Masson's trichrome to measure thickness of collagen bundles and hematoxylin-eosin (H&E) for histopathology. On Masson's trichrome, average diameter of 10 collagen bundles in every 10 fields were measured by digital lens Dino-eye, AM-7023, 5Mp and H&E sections for inflammatory cells and angiogenesis. Results suggested that Fraxinus excelsior 10% cream increased (P < 0.01) collagen bundle thickness in the papillary layer compared with eucerin but had no effect on collagen in the reticular layer. As expected, phenytoin increased (P < 0.01) collagenthickness in both reticular and papillary layers. .
