DNA Sequencing with DELTATaq DNA Polymerase

DNA sequencing using the dideoxy chain termination method of Sanger et al. (12) has proven to be quite effective in obtaining sequence information from a single-stranded DNA template. A denatured double-stranded DNA template can also be used to obtainDNA sequencing data (2,4,6,11) Improvements in DNA sequencing techniques are being introduced constantly. These improvements include the introduction of T7 DNA polymerase by Tabor and Richardson (13), which, with its high processivity, allows generation of sequence information of extended length. More recently, Taq DNA polymerase has found increasing use in DNA sequencing, especially when a limited amount of DNA template is available (7).
