The incorporation of radioactive isotopes into antibodies can combine the diagnostic and therapeutic possibilities of nuclear medicine with the exquisite selectivity of antibody targeting. Metal radionuclides can be tethered to antibodies by adding ligands that coordinate to the metal ion and preclude its release in vivo. Attaching the chelator to the antibody or fragment requires careful consideration to avoid compromising the affinity of the antigen-binding site for the target.
