Molecular mechanisms responsible for regulating gene expression in maturing grains of maize (Zea mays) and seedlings of wheat (Triticum aestivum) were analysed at the level of mRNA stability. The relative yield of poly(A)++mRNA (mRNA stability index) after the second cycle of poly(U)-sepharose chromatography was estimated. The in vitro incubation of aqueous preparations of total RNA led to differential degradation of mRNA similar to that observed in vivo under transcription inhibition by actinomycin D [dactinomycin]. The ranks of stability indices for seven specific mRNA from maturing maize grains and three mRNAs from wheat seedlings were determined. Comparison of stability indices for specific grain mRNAs of normal and opaque-2 maize showed thatthe stability index was the resulting value reflecting the correlation between stable and non-stable molecules in the mRNA pool.
