In Vitro Multiple Shoot Production in Sugarcane Genotypes

Shoot tips of improved sugarcane genotypes BO91, BO110, BO120 and BO128 were pretreated in a mixture solution of ascorbic acid citric acid, bavistin and streptomycin, surface sterilized and inoculated aseptically on MS basal medium supplemented with different concentrations and combinations of NAA and BAP. Out of the 15 different media combinations of NAA-BAP interaction, the establishment of expiant and growth of existing shoot was better in media with lower concentration of phytohormones particularly the BAP. Multiple shoots were formed from the swelled basal nodal region of the explant on media with either equal or higher concentration of cytokinin. Roots were differentiated on the in vitro developed shoots on a medium with 2.0 mg l~(-1) NAA and0.1 mg l~(-1) BAP. The plant-lets were acclimatized and transferred to the field. A micropropagation protocol was developed.
