Effect of media and explant on callus formation and plant regeneration in sorghum

Immature embryos and immature inflorescences of one sorghum genotype (2219B) were cultured on five culture media, viz. MS, B_5, SH, N_6 and MSB (vitamins of B_5 medium combined with mineral salts of MS medium) to compare the efficacy of explants andmedia. Callus induction frequency was not influenced by culture medium and explant. Callus growth in MS, B_5 and MSB media was comparable but better than that in N_6 and SH media. Shoot induction frequency on various media was comparable. Immature inflorescence-derived calli regenerated at a much higher frequency (51.9 percent) and produced more number of shoots/callus (1.4) than immature embryo-derived calli (39.5 percent and 0.9 respectively). Both explants produced maximum number of shoots on MSB medium but the number of shoots/callus from immature inflorescence-derived calli was almost twice (2.1) to that obtained from immature embryo-derived calli (1.1). Our results based on the genotype tested suggest that the two explants do not have tissue-specific requirement for culture medium and for better results MSB medium and immature inflorescences may be used for sorghum tissue culture.
