An important point when working with automated immunochemical techniques is to find suitable diluents for high-concentration samples. Although current immunometric techniques give a very wide analytical range and in many clinical situations sample dilutions are not necessary (1), to avoid interferences, the matrix of any diluents used should be as close as possible to that of normal sera from patients. Moreover, matrix problems vary from one sample to another and different immunoassays have different matrix problems (2). Diluents supplied by manufacturers in reagent kits usually do not show the same physicochemical properties as those of human serum, probably for legal or economic reasons. This means that clinical biochemists may have to assume that results obtained for high-concentration samples of different analytes may be erroneous or to prepare a suitable diluent to avoid these mistakes.
