The effects of plant growth regulators (NAA, 2,4-D, kinetin, BA or thidiazuron), sucrose (2, 4, 6 or 8%), light (0, 16 or 24 h light/24 h) and media (MS, NH, N6 or B5) on the production of callus and shoots of H. syriacus and on plant regeneration were investigated. Callus induction and growth was promoted in the presence of 2% sucrose and 16 h light. The best medium for callus induction was half-strength MS. Auxins promoted callus formation and cytokinins inhibited callus formation; 2,4-D (2-10 mg/litre) was more effective than NAA. Shoots were only induced in the presence of cytokinins and the absence of auxins. Good shoot formation was observed in the presence of kinetin at 1 mg/litre or BA at 0.5 mg/litre. Shoots also regenerated from callus in the presence of thidiazuron (0.1 or 0.5 mg/litre). Roots were produced following transfer to plant growth regulator-free media.
