Regenerative medicine has the potential to revolutionize transplantation through the generation of autologous replacement organs and tissues (i). Human induced pluripotent stem cells (iPSCs) have provided a powerful tool for the generation of cells of multiple lineages, however, a number of technical challenges in the generation of iPSCs remain. iPSCs are produced through 'reprogramming', the ectopic expression of a defined set of pluripotencyrelated transcription factors. Classical methods utilize retroviral or lentiviral vectors which promote permanent genetic modification.
