The increasing complexity of active pharmaceutical ingredients (APIs) is a challenge for all manufacturing organizations. Due to time pressure and high drop out rates investment in process R&D is usually kept at a minimum level in early drug development phases. This is an opportunity for service providers offering manufacturing solutions including route scouting based on a broad integrated chemical and biotech toolbox. Three examples of second generation processes are discussed where route scouting and combined chemistry and biotech capabilities were key: 1) resolution process based on a unique non-animal derived porcine liver esterase, 2) C-C-bond formation based on an evolved aldolase and 3) simultaneous C=C and C=O reductions based on a unique alcohol dehydrogenase and an enone reductase.
