We report a simple visual colorimetric method for the highly sensitive and selective detection of thiamine (Vitamin B1 ) by silver nanoparticles (AgNPs) as a probe. AgNPs surface was modified by glutathione (GSH) and their morphology, size distribution and optical properties were characterized by high res olution transmission electron microscope (HR-TEM) and various analytical spectroscopic techniques. The sensitivity of colorimetric assay is based on the surface plasmon resonance band (SPR) changes and aggregation of glutathione modified AgNPs (GSH-AgNPs) upon the successive addition of Vitamin B1. The selectivity of GSH-AgNPs based determination of Vitamin B1 is excellent, compared with other relevant vitamin interferences. GSH-AgNPs based sensor capably and specifically identified for the detection of Vitamin B1, it caused by their ability to form strong electrostatic interaction between positively charged Vitamin B1 and negatively charged GSH coated AgNPs. This spectral and visual colorimetric approach illustrates better sensitivity for Vitamin Bl with a lowest detection limit reaches 50.00 x 10~(-9) moldm~(-3). To establish the practical applications of the GSH-AgNPs probe in real sample analysis, the colorimetric assay was effectively applied to the detection of Vitamin B1 in spiked human blood serum and urine samples.
