Transgenic plants at a frequency of about 2.5% in an important crop, oilseed rape (Brassica napus), have been obtained through Agrobacterium-mediated transformation of cotyledon-petiole explants using two cosmid-type binary vectors (pOCA18 and pOCA28)that carry a kanamycin resistant gene (NPTII). It was observed that two Agrobacterium strains, an agropine-type strain (EHA105) and a nopaline-type strain (GV3101) and two cosmid vectors (pOCA18 and pOCA28) can be used to obtain transgenic plants. The integration of T-DNA in the B. napus genome was confirmed by Southern blot analysis, and the data suggested that there were one to four copies of T-DNA per integrated T-DNA locus.
