Solid phase microextraction (SPME) has been broadly used for sample purification and sensitivity enhancement, by eliminating interference compo-nents and reducing matrix effects. In this study, a method for preparing SPME column in 200 uL pi-pette tips using self-polymerization of dopamine. Solid extraction phase, graphite powder, was fixed by natural binder dopamine. Poly-dopamine link on the surface of the solid phase materials provide uni-form monolithic structure in extraction device. As both graphite powder and dopamine molecule pos-sess hydrophilic surface, the obtained SPME tips could retain the physical and chemical activity of the hydrophilic extraction and exhibit good extraction performance for carbohydrate molecules. The ex-traction parameters for oligosaccharides and glycans were investigated and optimized based on extraction results monitored by capillary electrophoresis analy-sis. This technique shows advantages like simple, economical, non-toxic and environmentally friendly.
