RhoGTPaseanditsupstreamactivator,guaninenucleotideexchangefactor2(RhoGEF2),haveemergedaskeyregulatorsofactinrearrangementsduringepithelialfoldingandinvagination(Nikolaidou,K.K.,Barrett,K.(2004).ARho-GTPase-signalingpathwayisusedreiterativelyinepithelialfoldingandpotentiallyselectstheoutcomeofRhoactivation.emCurr.Biol/em.14,1822ndash;1826).Here,weshowthatemDrosophila/em18wheeler(18W),aToll-likereceptorprotein,isanovelcomponentoftheRho-signalingpathwayinvolvedinepithelialmorphogenesis.em18w/emMutantembryoshavesalivaryglandinvaginationdefectssimilartoembryosthatlackcomponentsoftheRhopathway,andubiquitousexpressionof18WresultsinanupregulationofRhosignaling.Transheterozygousgeneticinteractionsanddoublemutantanalysissuggestthat18WaffectstheRho-GTPase-signalingpathwaynotthroughFogandRhoGEF2,butratherbyinhibitingRhoGTPaseactivatingproteins(RhoGAPs).WeshowthatRhoGAP5AandRhoGAP88C/Crossveinless-c(CV-C)arerequiredforpropersalivaryglandmorphogenesis,implicatingthemaspotentialtargetsof18W./p/div
