Thenuclearlaminaisanextensiveproteinnetworkthatunderliestheinnernuclearenvelope.Thisnetworkincludesthespanclass="underline"L/spanAP2-spanclass="underline"e/spanmerin-spanclass="underline"M/spanAN1-spanclass="underline"d/spanomain(LEM-D)proteinfamily,proteinsthatshareanassociationwiththechromatinbindingproteinBarrier-to-autointegrationfactor(BAF).LossofindividualLEM-Dproteinscausesprogressive,tissue-restricteddiseases,knownaslaminopathies.Mechanismsassociatedwithlaminopathiesarenotyetunderstood.HerewepresentourstudiesofoneoftheDrosophilanuclearlaminaLEM-Dproteins,Otefin(Ote),ahomologueofemerin.PreviousstudieshaveshownthatOteisautonomouslyrequiredforthesurvivaloffemalegermlinestemcells(GSCs).WedemonstratethatOteisalsorequiredforsurvivalofsomaticcellsintheovarianniche,withlossofOtecausingadecreaseincapcellnumberandalteredsignaltransduction.Weshowgermcell-restrictedexpressionofOterescuesthesedefects,revealinganon-autonomousfunctionforOteinnichemaintenanceandemphasizingthatGSCscontributetothemaintenanceoftheirownniches.Further,weinvestigatetherequirementofOteinthemalefertility.Weshowthatemote/emmutantmalesbecomeprematurelysterileastheyage.Paralleltoobservationsinfemales,thissterilityisassociatedwithGSClossandchangesinsomaticcellsoftheniche,phenotypesthatarelargelyrescuedbygermcell-restrictedOteexpression.Takentogether,ourstudiesdemonstratethatOteisrequiredautonomouslyforsurvivaloftwostemcellpopulations,aswellasnon-autonomouslyformaintenanceoftwosomaticniches.Finally,ourdataaddtogrowingevidencethatLEM-Dproteinshavecriticalrolesinstemcellsurvivalandtissuehomeostasis./p/div
