Previousstudieshavedemonstratedthatheadactivator(HA),an11aminoacidpeptide,stimulateshead-specificdifferentiationprocessesinhydra.Additionally,HAenhancesthedifferentiationofinterstitialcellsintonervecells.ThisstudyinvestigatedtheeffectsofexogenoussyntheticHAonthedynamicsofbothapicalandbasalregenerationinapieceoftissueexcisedfromthebodycolumnoftreatedanimalswhichcomprisedone-eighthoftheoriginalanimal.ThedynamicsofapicalandbasalregenerationweremonitoredusingthemonoclonalantibodyTS19.Thisantibodybindstoapicalandbasalectodermaltissueveryearlyintheprocessofregeneration,beforemorphologicalstructuresareevident.Labelingisultimatelylocalizedtothetentaclesoftheheadandaringabovethebasaldisc.Thus,TS19isausefultoolforanalyzingthedynamicsofbothapicalandbasalpatterningprocessesinthesameregeneratesimultaneously.QuantificationofTS19positiveareasonregeneratesoveratimecourseof72hrrevealedthatHAtreatmentacceleratedandamplifiedthedynamicsofbothapicalandbasalTS19labeling.Thespecificbasaleffectwasnovelandwasdemonstratedtooccurintheabsenceofadeterminedheadindependentlyofnewnervecelldifferentiation.Itisproposedthatthebasaleffectwastheresultofgrowthfactor-likeactivityofHA.
