emDrosophila/emDecapentaplegic(Dpp),amemberoftheBMP2/4classoftheTGF-βs,isrequiredfororgangrowth,patterninganddifferentiation.However,muchremainstobeunderstoodaboutthemechanismsactingdownstreamofthesemultipleroles.HereweinvestigatethisissueduringthedevelopmentoftheemDrosophila/emeye.Wehavepreviouslyidentifiedemviriato(vito)/emasadMyc-targetgeneencodinganucleolarproteinthatisrequiredforpropertissuegrowthinthedevelopingeye.Bycarryingoutatargetedeminvivo/emdouble-RNAiscreentoidentifygenesandpathwaysfunctioningwithVitoduringeyedevelopment,wefoundastronggeneticinteractionbetweenemvito/emandmembersoftheDppsignalingpathwayincludingtheTGF-βreceptorsemtkv/em(typeI),emput/em(typeII),andtheco-Smademmedea/em(emmed/em).AnalyzingtheexpressionoftheDppreceptorTkvandtheactivationpatternofthepathwayrsquo;stransducer,p-Mad,wefoundthatemvito/emisrequiredforacorrectsignaltransductioninDpp-receivingcells.Overall,wevalidatetheuseofdoubleRNAitofindspecificgeneticinteractionsand,inparticular,weuncoveralinkbetweentheDpppathwayandVito,anucleolarcomponent.emvito/emwouldactgeneticallydownstreamofDpp,playinganimportantroleinmaintainingasufficientlevelofDppactivityforthepromotionofeyediscgrowthandregulationofphotoreceptordifferentiationineyedevelopment./p/div
