Germlinestemcells(GSCs)inemDrosophila/emaredescendantsofprimordialgermcells(PGCs)specifiedduringembryogenesis.TheprecisetimingofGSCestablishmentinthetestishasnotbeendetermined,norisitknownwhethermechanismsthatcontrolGSCmaintenanceintheadultareinvolvedinGSCestablishment.Here,wedeterminethatPGCsinthedevelopingmalegonadfirstbecomeGSCsattheembryotolarvaltransition.Thiscoincideswithformationoftheembryonichub;thecriticalsignalingcenterthatregulatesadultGSCbehaviorwithinthestemcellmicroenvironment(niche).WefindthattheJakndash;STATsignalingpathwayisactivatedinasubsetofPGCsthatassociatewiththenewly-formedembryonichub.ThesePGCsexpressGSCmarkersandfunctionlikeGSCs,whilePGCsthatdonotassociatewiththehubbegintodifferentiate.IntheabsenceofJakndash;STATactivation,PGCsadjacenttothehubfailtoexhibitthecharacteristicsofGSCs,whileectopicactivationoftheJakndash;STATpathwaypreventsdifferentiation.Thesefindingsshowthatstemcellformationiscloselylinkedtodevelopmentofthestemcellniche,andsuggestthatJakndash;STATsignalingisrequiredforinitialestablishmentoftheGSCpopulationindevelopingtestes./p/div
