Wehaveusedquick-freezing,freeze-fracture,anddeepetchingtostudypostfertilizationgrowthofmicrovilliintheseaurchin,Strongylocentrotuspurpuratus.Between1and2minpostinsemination,weobservedrapidgrowthofnumerousfinger-likemicrovillitoabout0.7??minlength.Subsequently,groupsoftwotofourmicrovillibecameraisedofftheeggsurfaceonbroadcytoplasmicbases.Finally,by5minpostinsemination,thesebaseshadbecomeinterconnectedbythinsheetsofcytoplasmtoformacontinuousnetworkofrufflesacrosstheeggsurface.Correspondingthinsectionsofquick-frozenandfreeze-substitutedeggsshowedthatthesemicrovillicontainednetworksofmicrofilaments.Thefinger-likeprocesseshadbundlesofparallelmicrofilamentsthatrantheirentirelength,whiletherufflesofcytoplasmcontainedsheetsofcrisscrossedmicrofilaments.Theseobservationssupportthehypothesisthatmicrovillargrowthresultsfromformationofnewmicrofilamentnetworks.
