Corticesofunfertilizedseaurchineggscanbeisolatedinsuspensionandwilldischargetheattachedcorticalvesicles(CVs)inresponsetocalcium.WedescribeasimpleturbidometricassayformonitoringtheCa2+-induceddischargeofthesevesiclesandalsocomparethedischargeofvesiclesisolatedinahighsaltmedium(primarilyKCl)withamediummorecloselysimulatingtheinternalmilieuofthecell(primarilypotassiumgluconateandglycine).Dischargeinresponsetocalciumissimilarinbothmedia,requiringapproximately6??Mcalciumforone-halfmaximaldischarge.Thereare,however,significantdifferencesinmorphologyandproteincompositionofthetwotypesofpreparations(moreproteinspresentintheglycinecortices)andalsointherateofdischargeofthevesiclesinresponsetocalcium(KClcorticeswitht126secasopposedto30secintheglycinecortices).TheglycinecorticesgraduallylosetheirabilitytorespondtocalciumbutretentionofcalciumsensitivityisconsiderablyaidedbyinclusionofATPinthemedia;ATPhasnoapparenteffectondischargeoftheKClcortices.Theglycinecortices,asopposedtotheKClcortices,exhibitedvariationincalciumsensitivityduringthebreedingseasonandinthenumberofvesicleswhichwouldnotbreakdowninresponsetoaddedcalcium(referredtoasrefractoryvesicles).Thequestionofwhichtypeofcortexpreparationmostcloselysimulatestheinvivosituationisdiscussed,andtheviewispresentedthattheglycinecorticesmostcloselyresembletheinvivosituation.
