TheaccumulationofglobinmRNAinerythroidcellsduringhumandevelopmentwasinvestigatedinclonalculturesoffetal,newborn,andadultoriginerythroidprogenitorsandincontrolunculturederythroblastsandreticulocytes.Therelativeconcentrationsof?±,?2,and?3mRNAsequencesweremeasuredbyhybridizationwithpurified?±,?2,and?3[3H]cDNAprobes.Wefoundthattheamountof?3mRNAischaracteristicofthedevelopmentalstagefromwhichtheerythroidprogenitorcellsareobtained.Incoloniesfromfetallivercells,89%ofthenon-?±mRNAis?3mRNA.Coloniesderivedfromneonatal(cordblood)cellsproduceanaverageof55%?3mRNA,whilecoloniesgeneratedfromeitheradultbonemarroworprogenitorscirculatingintheperipheralbloodoftheadultindividualproduce?3mRNArangingfrom15to38%ofthenon-?±sequences.Whencomparedwitherythroidcells(erythroblastsorreticulocytes)fromthesametissues,thefetallivercoloniesproduceidenticalproportionsof?3mRNAaslivercellsinvivo.Coloniesfromcirculatingerythroidprogenitorsfromtheperinatal(switchover)periodaccumulatesomewhatless?3mRNA(55%)thandocirculatingreticulocytesinvivo(64%).Theconcentrationof?3mRNA(15to38%)incoloniesofadultmarroworperipheralbloodoriginissubstantiallyhigherthanthetraceamounts(&1%)foundinmatureerythroidcellsinvivo.Theamountsof?2and?3globinchainsynthesisfromfetalliver,cordblood,andadultperipheralbloodareproportionalto?2and?3mRNA(r=0.97)andthusindicatethatselectivetranslationof?2and?3mRNAisnotasignificantfactorintheswitchfromHbFtoHbAproduction.(However,bothneonatalreticulocytesinvivoanderythroidcoloniesinvitrocontainexcessnon-?±mRNAbutexhibitbalancedglobinchainsynthesis;translationalcontrolmechanismsapparentlypreventexcesssynthesisoftotalnon-?±chains.)Theexpressionofstage-specificpatternsof?3and?2mRNAinerythroidcoloniessuggeststhattheerythroidprogenitorcellsinvivopossessthepotentialforuniqueprogramsof?3and?2mRNAaccumulationbeforetheyactuallyenterthepathwaytoterminalmaturation.
