MixingofstarvedcellsofcomplementarymatingtypesofTetrahymenathermophiliainducesshorteningoftheirlongitudinallengthwithin10minofmixing.Thisearlymorphogenetictransformationinpreconjugantsexualinteraction(costimulationperiod)wasnamed"costimulation-inducedrounding"(CIR).CIRistheearliestmorphologicalchangethathaseverbeenfoundinthecostimulationperiodanddiffersfrom"synchronousrounding"inthevegetativecellcycle,becauseCIRcellsarestillabletoformfoodvacuoles,whilecellsinsynchronousroundingdonothavethisability.Whensexualcell-to-cellcollisionsbetweentwomatingtypeswerehamperedbyunidirectionalstirringfor20minaftermixingofthetwomatingtypes,bothCIRandconjugationweredelayedby20min.Whensecretedmaterialsneededfortheonsetofcostimulationwereremovedbywashingthecellswith10mMTris-HCl,pH7.4,beforemixingthetwomatingtypes,bothCIRandconjugationweredelayedbyabout30min.CIR-likeroundingwasnotinducedbycell-freemediumeitherfromtheoppositematingtypeorfrommixedcostimulatedcells.TheseresultsindicatethatCIRisinducedwhencellsareactivatedtoformconjugatingpairsbycell-to-cellcollisionsbetweencomplementarymatingtypesinthepresenceofsecretedmolecules.
