Neural influence on the expression of acetylcholinesterase molecular forms in fast and slow rabbit skeletal muscles☆

Withtheaimofinvestigatingtherolesofmotorinnervationandactivityonmusclecharacteristics,westudiedthemolecularformsofacetylcholinesterase(AChE)infast-twitch(semimembranosusaccessorius;SMa)andslow-twitch(semimembranosusproprius;SMp)musclesoftherabbit.WehaveshownthatSMaandSMpexpressdifferentpatternsandtissuedistributionofAChEformsandthattheeffectoflongdenervationvarieswithage.ThreeprincipalfindingsconcerningexpressionofAChEmolecularformsemergefromthesestudies.(1)TheactivityofAChEandthepatternofitsmolecularformsareparticularlyalteredinadultdenervatedSMaandSMpmuscles.AChEactivityincreasesby10-foldinbothmuscles,butasymmetricformsdisappearinSMaandincreaseby20-foldinSMpmuscles.AsimilaralterationofAChEisfoundaftertenotomyofthesemuscles,showingthattheeffectofdenervationmaybepartlyduetosuppressionofmuscleactivity.(2)ThedifferentchangesoccuringinthecompositionofAChEmolecularformsinadultdenervatedSMaandSMpmusclesareconsistentwithfluorescentstainingwithanti-AChEmonoclonalantibodiesandwithDBAorVVAlectins,whichbindtoAChEasymmetric,collagen-tailedforms.TheselectinspoorlystaindenervatedSMamusclesurfacesbutintenselystainneuromuscularjunctionsandextrasynapticareasindenervatedSMpmuscle.(3)Incontrastwiththeadult,denervationof1-day-oldmusclesdoesnotmarkedlymodifythetotalamountofAChEortheproportionsofitsmolecularforms,despitedramaticeffectsonmusclestructure.TheseresultsaresupportedbystudiesoflabelingwithfluorescentDBA:thelectinonlyslightlystainsthemusclefibersurfaceofdenervated15-day-oldSMpmuscle.Takentogether,thesedatashowthatdenervatedmusclesescapephysiologicalregulation,producingincreasedlevelsofAChEwithhighlyvariablecellulardistributionandpatternsofmolecularforms,dependingontheageofoperationandonthetypeofmuscle.
