Crystallin synthesis in lens differentiation in cultures of neural retinal cells of chick embryos☆

Dissociatedcellsofneuralretinasof3.5-day-oldchickembryosdifferentiatedintoa€?lentoidbodiesa€?withinabout10a€“12dayswhenculturedinvitro.ProteinsynthesisoftheseculturedcellswasstudiedwiththeuseofSDS-polyacrylamidegelelectrophoresis,affinitychromatography,andautoradiographycombinedwithimmunologicaltechniques.Incorporationof[14C]leucineintototalproteins,?±-crystallin,and?′-crystallinwasestimatedafterincreasingtimesofcultureuptoabout30days.Isotopeincorporationinto?′-crystallinwasdetectedat9days,anditincreasedsevenfoldafteranother17days.?±-Crystallinwasalsofirstdetectedat9days,butitsrelativecontentreachedamaximumat12daysandthendecreasedgradually.Theratioof?′-crystallinsynthesistototalproteinsynthesisincreasedupto40%at26days,whilethatof?±-crystallinsynthesisremained3%throughoutthecultureperiod.Theseresultsshowthatlensdifferentiationfromneuralretinalcellsisassociatedwiththepreferentialsynthesisoflenscrystallins,particularlyof?′-crystallin.
