PrimarymesenchymecellsusedinthisstudywereisolatedfromLytechinuspictusmesenchymeblastulaebytheirabilitytopreferentiallyadheretothesurfaceofatissueculturedishinthepresenceofserum.Onceisolated,primarymesenchymecellswerefoundtoformthin,elongated,activefilopodiawhichcloselyresemblethefilopodiaseeninvivo.Thefilopodiaformedinvitrocanmoveasstiffenedbristles,bendgraduallyorverysharply,orbeslowlywithdrawn.TheintegrityofthefilopodiaisnotaffectedbynocodazolebutistotallydisruptedbycytochalasinD.Filopodiaexhibitseveralapparentfunctionsinvitro:asorganellesinvolvedincontactingtheexternalenvironment,asanchoringappendagesthatholdthecellbodiesinplace,andasintercellularconnectivesthatcanjoincellbodies.Thefilopodiaofprimarymesenchymecellsappeartohavesimilarroleswithintheembryo.Thefunctionofthefilopodiahasbeenexploredbywatchingthebehaviorofisolatedprimarymesenchymecellsincloseproximitytodepositsofextracellularmaterial(ECM)preparedfrommesenchymeblastulae.WhenthefilopodiumfromamesenchymecellmakescontactwiththenearbyECM,aresponseisinitiatedwhichcausesthecellbodytomoveinadirectedmannertowardtheECMdeposit.Theuseofthistypeofresponseasamodelsystemforthestudyofthemigrationofprimarymesenchymecellswithintheembryoisconsidered.
