Characterization of antisera against mouse teratocarcinoma OTT6050: Molecular species recognized on embryoid bodies, preimplantation embryos, and sperm

AntiseraraisedinrabbitsbyhyperimmunizationwithsmallembryoidbodiesofthetransplantableteratocarcinomaOTT6050recognizeseveraldistinctantigenicproteinspeciesonthesurfacesofcellsoftheimmunogen.Someoftheseantigenswerefoundonthecellsofpreimplantationmouseembryos,oncellsfromparietalyolksaccarcinoma,andonmousesperm.Theseantigenshavebeendistinguishedbypolyacrylamideelectrophoresisoftheimmuneprecipitatesfromdetergentextractsoflactoperoxidase-iodinatedcells.TheintactembryoidbodiesfromtheasciticformoftheOTT6050teratocarcinomaexhibitedfivemajorproteinbands(approximateMW150K,115K,82K,48K,and12K),onebandthatranatthedyefrontofthegels(Rfa‰￥1)andoneminorband(approximateMW22K).Twodifferentrabbitantiserarecognizedanessentiallyidenticalpatternofantigenswhich,however,variedonthedifferentcelltypestested.Antiserawerealsoelicitedinsyngeneicmalemiceusingglutaraldehyde-fixedorirradiatedOTT6050embryoidbodies.Theisoantiserahadverypoortitersincomparisontotheabsorbedxenoantisera,asassessedbycomplement-mediatedcytotoxicactivityagainsttheimmunizingcelltypes.Complement-mediatedcytotoxicitycouldalsobedemonstratedusingparietalyolksaccarcinomacells,preimplantationmouseembryosfromallcleavagestages,blastocysts,andimmunosurgicallyisolatedinnercellmasses,astargets.Thecomplexityoftheantiserageneratedbyintactembryoidbodiesdescribedhereindicatesthatthesestructuresbearmultipleantigenicspecificitiesnotpresentonadultsomaticcells,someofwhicharestage-specificembryonicpolypeptides.
