Thedifferentiatedphenotypeofrabbitarticularchondrocytescanbecharacterizedbythesynthesisofhighlevelsofcartilagespecificproteoglycanandcollagen(typeII).Treatmentofthesecellsinprimarymonolayercultureforperiodsofupto18dayswith0.03to3.0??g/mlretinioicacid(RA)resultedinsuppressionofcolonyformation,alteredmorphology,anddecreased(eightfold)proteoglycanandcollagensynthesis.Withtheexceptionofcollagensynthesis,thesechangeswerecompletewithalldosesafter4daysoftreatment.Collagensynthesisdeclinedmoreslowly;itwasdosedependentafter4daysandmaximallyinhibitedbyalldosesby9days.DetailedanalysisofthecollagenphenotypewasperformedusingSDS-PAGEofintactchainsand2-DCNBrpeptideanalysis.RAcausedcessationoftypeIIsynthesis,andtransientstimulationoftypeIIIandtypeItrimercollagensynthesis,withoutinductionoftypeIcollagen.Essentiallyidenticalresultswereobtainedwithretinol.TheresultantcollagenphenotypedifferedsignificantlyfromthetypeI-containingphenotypeinducedbysubculture.Thus,suppressionofthisdifferentiatedprogramdidnotelicitacommonmodulatedphenotype.TheresultsarediscussedinthecontextofdirectandindirectmechanismsofRA-dependentmodulationofchondrocytegeneexpression.
