MitochondrialATPsynthaseisamotorenzymeinwhichacentralshaftrotatesinthestatorcasingsfixedwiththeperipheralstatorstalk.Whenexpressionofd‐subunit,astatorstalkcomponent,wasknocked‐down,humancellscouldnotformATPsynthaseholocomplexandinsteadaccumulatedtwosubcomplexes,onecontainingacentralrotorshaftpluscatalyticsubunits(F1‐c‐ring)andtheothercontainingstatorstalkcomponents(“b–e–g”complex).F1‐c‐ringwasalsoformedwhenexpressionofmitochondrialDNA‐codeda‐subunitandA6Lwassuppressed.Thus,thecentralrotorshaftandthestatorstalkareformedseparatelyandtheyassemblelater.SimilarassemblystrategyhasbeenknownforATPsynthaseofyeastandEscherichiacoliandcouldbecommontoallorganisms...
