TheknownfunctionsoftypeIIthioesterases(TEIIs)intypeIpolyketidesynthases(PKSs)includeselectingofstarteracylunits,removalofaberrantextenderacylunits,releasingoffinalproducts,anddehydrationofpolyketideintermediates.Inthisstudy,wecharacterizedtwoTEIIs(ScnIandPKSIaTEII)fromStreptomyceschattanoogensisL10.DeletionofscnIinS.chattanoogensisL10decreasedthenatamycinproductionbyabout43%.BothScnIandPKSIaTEIIcouldremoveacylunitsfromtheacylcarrierproteins(ACPs)involvedinthenatamycinbiosynthesis.OurresultsshowthattheTEIIcouldplayimportantrolesinboththeinitiationstepandtheelongationstepsofapolyketidebiosynthesis;theintracellularTEIIsinvolvedindifferentbiosyntheticpathwayscouldcomplementeachother...
