Cell-based therapy of type 1 diabetes mellitusresides on beta cell replacement and isletregeneration strategies. Various cell types ofembryonic, foetal and adult origin have beeninvestigated. So far, the most adequatealternative cell source is still to be defined.Human islet pancreatic derived cells (hIPC)have been demonstrated to differentiate invitro into beta cell phenotypes. In previouswork we have shown that stromal hIPCsdisplay mesenchymal stem cell features. Here,we compared the stemness character of hIPCsand human bone-marrow derivedmesenchymal stem cells (hMSC) as well asthe in vitro potential of these two cellpopulations to differentiate into insulinproducing cells.Materials and methods:Gene expression pattern was analysed inhIPCs and hMSC by means of cDNAmicroarray hybridization (Affymetrix U133Plus 2.0 chip) and stem cell marker geneswere confirmed by RT-PCR. hIPCs and hMSCwere expanded and cultivated in specificpancreatic endocrine promoting conditions(high glucose serum free medium, Activin-A,s-cellulin, hHGF, exendin-4 and nicotinamid).After 5 days in differentiation medium, isletgene markers were investigated by means ofRT-PCR and immunocytochemistry in bothcell populations
