Backgroundandobjective:Thecell-surfacelipooligosaccharide(LOS)ofTreponemadenticolapossessesseveralbiologicalproperties.TheaimofthisstudywastoinvestigatethebindingpropertiesofT.denticolaLOStoextracellularmatrix(ECM)proteins,mucosalcells,andoralbacteria.Design:LOSwasisolatedfromT.denticolaandlabeledwithtritium.Tritium-labeledLOSwasplacedinECMprotein-,epithelialcell-,fibroblast-,orbacterium-coatedwellsofa96-wellmicroplate.Followingincubation,unattachedLOSwasremovedbyextensivewashing,andtheamountofboundLOSwasdeterminedbymeasuringtheradioactivityinthewells.PeptostreptococcusmicroscoatedwithLOSwasusedtostimulatefibroblasts,andthesecretionofinterleukin-6(IL-6)andinterleukin-8(IL-8)bythefibroblastswasdeterminedbyELISA.Results:T.denticolaLOShadahighaffinityforlaminin.Italsoboundtogingivalepithelialcellsandfibroblasts.SolubleCD14significantlyincreasedthebindingofLOStofibroblasts.MoreLOSboundtoP.microsthantheotheroralbacterialspeciestested.StimulatingfibroblastswithLOS-coatedP.microsinducedthesecretionofIL-6andIL-8.Conclusions:OurstudyprovidedevidencethatT.denticolaLOSpossessesthecapacitytobindtoECMproteins,mucosalcells,andoralbacteria.Inaddition,LOSbindingtobacteriamayincreasetheirpro-inflammatorypotential.?Keywords:adherence;cytokine;epithelialcells;fibroblasts;laminin;lipooligosaccharide;periodontaldisease;Peptostreptococcusmicros;Treponemadenticola(Published:16September2013)Citation:JournalofOralMicrobiology2013,5:21517-http://dx.doi.org/10.3402/jom.v5i0.21517
