Genome‐wideassociationstudieshaveprovidedmanygeneticalterations,conferringsusceptibilitytomultifactorialpolygenicdiseases,suchasinflammatoryboweldiseases.Yet,howspecificgeneticalterationsfunctionallyaffectintestinalinflammationoftenremainselusive.Itisnoteworthythatalargeoverlapofgenesinvolvedinimmunedeficiencieswiththoseconferringinflammatoryboweldiseaseriskhasbeennoted.Thishasprovidednewargumentsforthedebateonwhetherinflammatoryboweldiseasearisesfromeitheranexcessoradeficiencyintheimmunesystem.Inthisreview,wehighlightthefunctionaleffectofaninflammatoryboweldisease‐riskallele,whichcannotbededucedfromgenome‐wideassociationstudiesdataalone.Asexemplifiedbythetranscriptionfactorsignaltransducerandactivatoroftranscription3(STAT3),weshowthatasinglegenecanhaveaplethoraofeffectsinvariouscelltypesofthegut.Theseeffectsmayindividuallycontributetotherestorationofintestinalhomeostasisontheonehandorpavethewayforexcessiveimmunopathologyontheother,asaninflammatory“rheo‐STAT”...
