TheUVB(290–320nm)radiationinsunlightisresponsibleforinducingskincancer.ExposuretoUVradiationisalsoimmunosuppressive,andthesystemicimmunesuppressioninducedbyUVisawell‐recognizedriskfactorforcancerinduction.AsUVBradiationisabsorbedwithintheupperlayersoftheskin,indirectmechanismsmustplayaroleinactivatingsystemicimmunesuppression.Oneprominentexampleismastcellmigration,whichfromtheskintothedrainingLNisanessentialstepinthecascadeofeventsleadingtoimmunesuppression.Whattriggersmastcellmigrationisnotentirelyclear.Here,wetestedthehypothesisthatPAF,alipidmediatorofinflammationproducedbytheskininresponsetoUVexposure,isinvolved.Mastcell‐deficientmice(KitW‐sh/W‐sh)areresistanttothesuppressiveeffectofUVradiation,andreconstitutingmastcell‐deficientmicewithnormalbonemarrow‐derivedmastcellsrestoressusceptibilitytoimmunosuppression.However,whenmastcellsfromPAFR?/?micewereused,thereconstitutedmicewerenotsusceptibletothesuppressiveeffectsofUV.Furthermore,PAFR?/?miceshowedimpairedUV‐inducedmastcellmigrationwhencomparedwithWTmice.Finally,injectingPAFintoWTmicemimickedtheeffectofUVirradiationandinducedmastcellmigrationbutnotinPAFR?/?mice.OurfindingsindicatethatPAFRbindinginducesmastcellstomigratefromtheskintotheLNs,wheretheymediateimmunesuppression...
