Genetic Variation within Glycine max (L.) Merrill Genotypes Resistant to Fusariose as Revealed by Microsatellite Markers

Fifteen soybean varieties, which are part of collection of the Institute of Genetics and Plant Physiology of the Academy of Sciences of Moldova, were analyzed by means of in vitro/in vivo Fusarium oxysporum resistance determination and by molecular techniques, using microsatellite (SSR) markers. The majority of the genotypes were newly developed and of a great value due to high levels of resistance to fusariose. A total of 19 SSRs, distributed on 19 of 20 linkage groups of soybean, were used to amplify specific microsatellite loci from bulked DNA of 15 genotypes. Out of 19 SSR loci tested 14 generated results and 12 were successful in distinguishing the cultivars resistant to fusariose. A total of 60 alleles were detected and used for genetic similarity estimations and genotype clustering. The number of alleles per locus varied from one to nine, with an average of 4.3 alleles per locus. Multiallelic microsatellite loci were revealed in all but two cases. The dendrogram based on the SSR loci profiles showed good agreement with the resistance characteristics of cultivars. The varieties with the highest levels of resistance were grouped together forming a single cluster.
