Aserologicalassaybasedonthedetectionofgoatpregnancyassociatedglycoprotein(caPAG)bythesandwich-ELISAmethodwasdesignedandcarriedoutonserumofAfricandwarfgoatinothertodevelopanearlypregnancydiagnosis.Forthispurpose,theanti-caPAGantibodiesproducedinrabbitswerebiotinylatedandtitratedforthedevelopmentoftheELISA"sandwich"assayforthecaPAGdetectioninthegoatserum.BloodsampleswerecollectedeverysevendaysfromsixAfricandwarfgoatsafterinduction,synchronizationofestrusandprojection.SeraobtainedhaveservedinserologicalassaybyELISA"Sandwich".Theopticaldensitiesreadat492nmonanELISAautomaticreaderhasservedtoestablishtheprofilesofthecaPAG.Pregnancyassociatedglycoprotein#8217;srateincreasesrapidlyduringthefirstthirdofpregnancytoapeakcorrespondingtoaconcentrationof117ng/mlatday91ofgestation.Thisconcentrationthendropstoreachabout50ng/mlthe121stdayandremainsmoreorlessstableuntilparturitionwhereitgraduallydropstobaselinevalues.However,threeweeksafterparturition,therateofPAGisstillhighinthematernalcirculation.GoatPAGisdetectableinthematernalcirculationofpregnantAfricandwarfgoatsbyELISA"sandwich"fromday28afterfertilizationwithsensitivityof100%.Betweendays14and21thesensitivitiesarelowandarerespectivelyof33.3%and66.7%.ThissandwichELISAgivesareliablelaboratorytestandconvenientalternativeforanearlypregnancydiagnosisfromday28afterinsemination.Itisthereforeamajorstepforwardforearlydiagnosisofpregnancyingoats.
