AnautomatedmultiplexPCRassayof10polymorphicmicrosatellitelociwasoptimizedhereinSenepolbovinecattle.AllelefrequencieswereestimatedforColombianherds.Thepowerofthissetofmarkersforparentageandkinshipanalyseswasalsotested.Populationgeneticparameters,combinedprobabilityofidentity(CPI)andcombinedprobabilitiesofexclusion(CPE)wereestimated.Theaveragenumberofallelesperlocuswas14,theeffectivenumberofalleleswas5,theobservedandexpectedheterozygositieswere0.635and0.776,respectively,theShannonIndexwas2.70andthepolymorphicinformationcontentwas0.751.Thesevaluesevidencedelevatedlevelsofpolymorphismandanunderestimationofgeneticdiversitythroughtheuseofmanualgenotypingmethods.Inbreedingcoefficient(Fis=0.206)inColombianSenepolherdswasmoderate.Altogether,datafromtheusedsetofmicrosatellitelociyieldedaCPIbetweentworandomsamplesof1.07E-12.TheCPEinparentagetestingwere0.998forthefirstparentand0.999forboththesecondparentandparentpairs.Thissetofpolymorphicmarkerscanbegenotypedinarelativelyeasy,effectiveandreliablewayforrelatednessandforensicanalyses.Finally,werecommendourapproachforpopulationgeneticanalysesofotherSenepolpopulationsandfuturegeneticmonitoringofinbreedinginSenepolherds.
