Serial Laparoscopic Biopsies of Liver and Spleen from Schistosoma-Infected Baboons (Papio spp.)

Purpose:Toobtainlarge,serialbiopsysamplesfromtheliverandspleenbyusinglaparoscopy.Largesampleswereneededformeasurementofinflammatorymediatorsduringvariousstagesofschistosomiasis.brxmlns="http://pub2web.metastore.ingenta.coms/"/brMethods:Eachofthesevenfemalebaboons(Papiosp.)underwentasmanyasthreelaparoscopies,foratotalof19laparoscopicprocedures.Thisprocesspermittedsamplingoftheliver,spleen,andmesentericlymphnodesbeforeandat6and9weeksafterinfectionwithSchistosomamansoni.Allsurgerywasperformedthroughthreetrocarsites.Postoperativecareincludedpreemptiveanalgesia.Aftersurgery,wemonitoredtheanimals'appetiteandmeasuredthecorebodytemperatureandactivitybyusingimplantedradiofrequencytransmitters.brxmlns="http://pub2web.metastore.ingenta.coms/"/brResults:Weobtainedsamplesoftheliverandsplenicbiopsiesduringall19laparoscopicprocedures.Themeanweightoftheliverbiopsieswas3.7gandthatofthespleensampleswas5.3g.Weencounteredsmalladhesionsduring5ofthe12reoperations.Eatingandactivityrapidlyreturnedaftersurgery.brxmlns="http://pub2web.metastore.ingenta.coms/"/brConclusions:Laparoscopypermittedcollectionoflarge,serialbiopsieswithapparentlylimitedstresstotheanimals.Laparoscopycanbeusedforbiopsiesinstudiestocharacterizediseaseresponse,confirmnormalorganhistologypriortodrugtoxicitystudies,determinetarget-organdrugconcentrationsinpharmacokineticstudies,andmeasuredrugresidues.Thisrefinementlikelywillreducerequiredanimalnumbersbydecreasingtheeffectofsurgerycomparedtothatoftheexperimentalconditions,enhanceanimalwell-being,andpermitrepeatedmeasurementsinananimalthatservesasitsowncontrol.
