Laboratorymiceroutinelyarehousedat20to22°C#8212;wellbelowthemurinethermoneutralzoneof29to34°C.Chroniccoldstressrequiresgreaterenergyexpendituretomaintaincorebodytemperatureandcanleadtothefailureofmousemodelstoemulatehumanphysiology.Wehypothesizedthatmicehousedatambienttemperaturesof20to22°Carechronicallycold-stressed,havegreaterenergyexpenditure,andhavehighglucoseutilizationinbrownadiposetissue.Totestourhypotheses,weusedindirectcalorimetrytomeasureenergyexpenditureandsubstrateutilizationinC57BL/6JandCrl:NU-Foxn1supnu/supnudemiceatroutinevivarium(21°C),intermediate(26°C),andheated(31°C)housingtemperatures.Wealsoexaminedtheactivationofinterscapularbrownadiposetissue,theprimarysiteofnonshiveringthermogenesis,viathermographyandglucoseuptakeinthisregionbyusingpositronemissiontomography.Energyexpenditureofmicewassignificantlyhigheratroutinevivariumtemperaturescomparedwithintermediateandheatedtemperaturesandwasassociatedwithashiftinmetabolismtowardglucoseutilization.Brownadiposetissueshowedsignificantactivationatroutinevivariumandintermediatetemperaturesinbothhirsuiteandnudemice.Crl:NU-Foxn1supnu/supmiceexperiencedgreatercoldstressthandidC57BL/6Jmice.Ourdataindicatemicehousedunderroutinevivariumconditionsarechronicallycoldstress.Thisnoveluseofthermographycanmeasurecoldstressinlaboratorymicehousedinvivaria,akeyadvantageoverclassicmetabolicmeasurementtools.Therefore,thermographyisanidealtooltoevaluatenovelhusbandrypracticesdesignedtoalleviatemurinecoldstress.
