Hypervitaminosis D in Guinea Pigs with α-Mannosidosis

Acolonyofguineapigs(n=9)with#945;-mannosidosiswasfedapelletedcommerciallaboratoryguineapigdiet.Over2mo,all9guineapigsunexpectedlyshowedanorexiaandweightloss(11.7%to30.0%ofbaselineweight),and3animalsdemonstratedtransientpolyuriaandpolydipsia.Bloodchemistrypanelsinthese3guineapigsrevealedhigh-normaltotalcalcium,high-normalphosphate,andhighALP.Urinespecificgravitywasdilute(1.003,1.009,1.013)inthe3animalstested.Postmortemexaminationof7animalsthatwereeuthanizedafterfailingtorespondtosupportivecarerevealedrenalinterstitialfibrosiswithtubularmineralization,softtissuemineralizationinmultipleorgans,hepaticlipidosis,andpneumonia.AnalysisofthepelleteddietrevealedthatithadbeenformulatedwithavitaminD3contentofmorethan150timesthenormalconcentration.Ionizedcalciumand25-hydroxyvitaminDvalueswerebothhighinserumsavedfrom2euthanizedanimals,confirmingthediagnosisofhypervitaminosisD.Thisreportdiscussestheclinicalsigns,bloodchemistryresults,andgrossandhistologicfindingsofhypervitaminosisDinacolonyofguineapigs.Whenunexpectedsignsoccurcolony-wide,dietarydifferentialsshouldbeinvestigatedatanearlytimepoint.
