Validation of an Enzyme-Linked Immunosorbent Assay Kit Using Herpesvirus Papio 2 (HVP2) Antigen for Detection of Herpesvirus Simiae (B virus) Infection in Rhesus Monkeys

SerologictestingforantibodytomonkeyBvirus(BV)inmacaqueseraisproblematicduetothebiohazardousnatureofBVantigens.Herpesviruspapio2(HVP2),aherpesvirusofbaboons,isnonpathogenictohumansandisgeneticallyandantigenicallymorecloselyrelatedtoBVthanishumanherpessimplexvirus1.Thispaperdescribestheresultsofourin-houselaboratorythatcomparedaBVantigen-basedenzyme-linkedimmunosorbentassay(ELISA)bycommercialtestinglaboratoryandanHVP2-basedELISAinourlaboratorybyusing447serafrom290rhesusmonkeys.TheHVP2-basedELISAidentifiedaspositive99.11%oftheseraidentifiedasBV-positivebytheBVELISA.TheBVantigen-basedELISAidentifiedaspositive98.21%oftheseraidentifiedasBV-positivebytheHVP2-basedELISA.TheHVP2ELISAalsoidentifiedtwoBV-negativeandsixBV-equivocalseraaspositive.BothELISAsidentifiedthesame85negativeandthreeequivocalsamplesasnegativeandequivocal,respectively.Thehighdegreeofcorrelation(weightedkappacoefficient,0.94)betweenthetwotestsindicatesthattheHVP2ELISAisasensitiveandreliableassayforin-housetestingoftheBVstatusofrhesusmonkeys.
