Melatonin(MLT),thecircadianneurohormonesecretedbythepinealglandinmammalsduringdarkness,eicosapentanoicacid(EPA),andconjugatedlinoleicacid(CLA)haveestablishedregulatoryrolesincancergrowth.Investigationsinourlaboratoryhaveindicatedthattheseagentsinhibitfattyacid(FA)transportbytumorsandseveralsub-typesofwhiteadiposetissueviainhibitoryGprotein-coupledreceptormechanisms.Skeletalmuscleconstitutesover45%ofhumanbodymassandplaysanimportantroleincancercachexiaandobesity-relateddiseases.Sincefattyacidoxidationisamajorsourceofenergyforthistissue,wetestedthehypothesisthatphysiologicMLTlevels,EPA,orCLAinjectedintravenously,inhibitFAuptakeinratskeletalmuscleinvivo.Weusedasurgicaltechniqueforcatheterizingthefemoralveininratsthatallowsrapidbloodcollectionfromtheentirehindlimb,whileensuringcontinuousbloodflowtothetissue.Bloodacid/gastensionsandhematocritweremonitoredandremainedconstantduringthecourseofeachexperiment.TheMLT,EPA,andCLAinhibitedFAuptakebythetissueandloweredcAMPvalues.Glucoseuptakeandglycerolproductioninthehindlimbwerenotaffected.TheseinvestigationssuggestanovelroleforMLT,omega-3FAs,andCLAintheregulationofFAtransportandfatmetabolisminskeletalmuscle.
