Thefrequentoccurrenceoffatigueanddisturbedsleepincancersurvivorsandthenegativeeffectofthesesymptomsonqualityoflifeandclinicaloutcomeunderscoretheneedtoidentifymechanismsthatcausecancer-relatedfatigue,withaviewtowarddevelopingmoreeffectivetreatmentsforthisproblem.Humanstudiesoffatigueanddisturbedsleeparelimitedbyhighinter-individualgeneticandenvironmentalvariability,difficultieswithbehavioralorreportingcompliance,andthesubjectivenatureoftheproblems.Althoughanimalmodelsalsomustovercomethebarrierofassessingfatigueandsleepdisturbanceintheabsenceofobviousobjectiveclinicalmarkers,animalstudiesareeasiertocontrolandstandardizethanarestudiesofpeople.Moreover,animalmodelsarecrucialtotheidentificationandunderstandingofunderlyingdiseasemechanisms.Thisreviewdescribestheneedfor,thefeasibilityof,andseveralpossibleapproachestomeasuringfatigueinanimalmodelsofcancerandtorelatingsuchmeasurestodisturbedsleep,immunefunction,andotherpotentialmechanisms.Developingandusinganimalmodelstobetterunderstandfatigueanddisturbedsleeprelatedtocanceranditstreatmenthasanenormouspotentialtoexpandtheknowledgebaseandfosterhypothesesnecessaryforthefuturedevelopmentandtestingofinterventions.
