Aortic Response to Balloon Injury in Obese Zucker Rats

Thesmalldiameterofthecarotidarteryisnotcompatiblewiththeevaluationofclinicallyavailableendovasculardevicesinthecarotidballoon-injury(BI)model.WedevelopedanendovascularBImodelintheratdescendingaorta,whosesizeiscompatiblewithavailableendovascularinstruments.WealsotestedthehypothesisthatneointimaformationisenhancedintheaortaofobeseZuckerrats(OZR)comparedwithleanZuckerrats(LZR).LeftexternalcarotidarteriotomiesandBIofthethoracicandabdominalaortawereperformedbyusingaballooncatheter.Aortogramsandaorticpathologywereexaminedat2,4,and10wkafterBI.At10wkafterBI,theabdominalaortainOZRhadnarrowed8.3%±1.1%relativetobaselinecomparedwithanexpansionof2.4%±2.2%inLZR.Simultaneously,thethoracicaortahadexpanded9.5%±4.3%inLZRcomparedwithstenosisof2.8%±1.6%inOZR.Calculationoftheintimal:medialthicknessratiorevealedsignificantlyincreasedneointimalformationintheOZRdescendingaortacomparedwiththatinLNR.Inconclusion,thisminimallyinvasiveBImodelinvolvingtheratdescendingaortaiscompatiblewithavailableendovascularinstruments.ThedescendingaortaofOZRdemonstratesenhancedneointimalformationandconstrictivevascularremodelingafterBI.
