AnoutbreakofmousepoxinaresearchinstitutionwascausedbyEctromelia-contaminatedmouseserumthathadbeenusedforbonemarrowcellcultureandthecellssubsequentlyinjectedintothefootpadsofmice.ThediseaseinitiallywasdiagnosedbyidentificationofgrossandmicroscopiclesionstypicalforEctromeliainfection,includingfociofnecrosisintheliverandspleenandeosinophilicintracytoplasmicinclusionbodiesintheskin.ThesourceofinfectionwasdeterminedbyPCRanalysistobeserumobtainedfromacommercialvendor.Todeterminewhetherviralgrowthintissueculturewasrequiredtoinduceviralinfection,36mice(BALB/cJ,C57BL/6J)wereexperimentallyexposedintraperitoneally,intradermally(footpad),orintranasallytocontaminatedserumorbonemarrowcellculturesusingthecontaminatedserumintheculturemedium.Micewereeuthanizedwhenclinicalsignsdevelopedorafter12wk.Necropsy,PCRofspleen,andserumELISAwereperformedonallmice.Miceinjectedwithcellculturesandtheircagecontactsdevelopedmousepox,antibodiestoEctromelia,andlesions,whereasmiceinjectedwithserumwithoutcellsdidnot.Mouseantibodyproduction,atoolcommonlyusedtoscreenbiologicmaterialsforviralcontamination,failedtodetectactiveEctromeliacontaminationinmouseserum.
